The Centers for Disease Control (CDC) / National Center for Immunization and Respiratory Diseases (NCIRD)/ Division of Bacterial Diseases (DBD), Respiratory Diseases Branch (RDB) intends to award a sole source firm fixed price Purchase Order to New England Biolabs of Ipswich, MA, for the acquisition of Bst 2.0 WarmStart DNA Polymerase. New England Biolabs is the only contractor that provides a strand-displacing polymerase with a reversibly-bound aptamer that inhibits polymerase activity below 45°C.The Bst 2.0 polymerase has an aptamer that reversibly binds to the enzyme, inhibiting activity below 45°C, i.e. during reaction assembly at room temperature. This binding event ensures that non-specific amplification events do not occur at permissive lower temperatures during set-up. Additionally, this enzyme enables rapid amplification of nucleic acid under isothermal conditions and is an essential component of point-of-care assays in development. In experiments, CDC researchers have shown that the Bst 2.0 form of the enzyme, designed in silico by New England Biolabs, amplifies target more rapidly and sensitively than other strand-displacing enzymes that do not possess the "warm start" feature.
