USDA, ARS, USHRL, intends to award a sole source purchase order to Rapid Genomics; Gainesville, FL for Hybridization-based capture of target regions, followed by next-generation sequencing. Both high-output and high-throughput genotyping is required.
Hybridization-based capture of target regions, followed by next-generation sequencing, is the desired strategy. Workflow is as follows:
1. Development of a set of 10000 validated probes for sugarcane, consisting of biotinylated 120-mer probes that complement a segment of each of the target sequences.
2. Submitted DNA samples (total of 400) will be processed to sequencing compatible libraries.
4. Probes will be utilized to capture sequences at each target locus, and captured targets will be sequenced using next-generation sequencing.
5. Upon sequencing, data will be analyzed through a bioinformatics pipeline to
generate SNP markers in the target probe region.
Results expected include approximately 10,000 SNP markers for each of 400 sugarcane genotypes, plus availability (if requested), of raw and/or minimally processed sequencing data.
Our justification to award is in accordance with Federal Acquisition Regulation (FAR) 6.302-1 Only one responsible source and no other supplies or services will satisfy agency requirements.
