A major research goal of the NIDDK to discover/develop small, "drug-like" molecules that are
agonists of the thyrotropin (thyroid-stimulating hormone, TSH) receptor (TSH-R) to be used as probes for TSH-R function. Under the Molecular Libraries Probe Production Centers Network (MLPCN) known drug or drug-like compounds were screened for activity at this receptor. The ultimate goal was to identify a lead compound for eventual use as a drug to diagnose and treat patients with thyroid cancer. Using a high-throughput screen, we discovered a successful probe that is a TSH-R agonist and have tested over 100 analogs of this active compound. Development of a lead drug candidate has progressed to the point that the lab now has an active pharmaceutical ingredient (API) that is a highly efficacious, highly potent TSH-R agonist in vitro. To be able to move this API forward for use in vivo in animal models and to obtain sufficient data to submit an application to the Food and Drug Administration for an investigational New Drug, we need to continue development.
It is anticipated that the actual work will take approximately 2 months from receipt of materials from NIDDK.
