The USDA, ARS, Western Regional Research Center, Albany, California has a requirement a refurbished benchtop protein kinetics/affinity measuring system for quantitation and kinetics using label-free, real-time analysis that can measure 8 samples in parallel for kinetics or concentration determination. This protein interaction system must enable measurement of kinetic constants (ka, kd, and KD) for protein:protein interactions. The system must also enable measurement of protein concentrations, as determined from a standard curve with known concentrations, including the capability to measure immunoglobulin G (IgG) concentrations within the range of 0.5 mcg/mL to 500 mcg/mL time within 2 minutes for 8 samples and 32 minutes for 96 samples. The concentration determination must be based on kinetic measurements, without the need for background subtraction. The system shall be suitable for use with crude samples and be non-destructive to the sample for full recovery and usage, without cross-contamination of samples. The system must be fluidics-free and be able to measure 8 samples in parallel for kinetics or concentration determination of samples in a standard 96-well microplate and the sensing element (biosensors) must be disposable with optional regeneration. The biosensors must be available with the following chemistries: amine-reactive, streptavidin, Protein A, anti-penta-HIS, and anti-mouse IgG. Data handling and storage must include an integrated software package and Windows-based PC and interfacing, with provision for data export as text files.
