Description of Use:
The isothermal titration calorimeter will be used to directly measure heat evolved or absorbed in liquid samples normally as a result of injecting precise amounts of reactants. The sample and a buffer reference solution are enclosed in an adiabatic jacket. During an experiment the reference cell is heated by a very small constant power, the reference offset. The temperature difference between the two cells is constantly measured and a proportional power is increased or reduced to the sample cell by the cell feedback system to keep the temperature difference very small. A signal proportional to that cell feedback power is called CFB, and with the instrument temperature and time, constitutes the instrument's relevant raw data.
Specific and immediate applications of the isothermal titration calorimetry:
I. Characterize the molecular interactions proposed to use ITC to fully characterize the thermodynamics of protein-protein and protein-RNA interactions that are implicated in the biological function of various biomolecules. In particular, interactions mediated by virion infectivity factor (Vif), which include those with RNA and cellular proteins.
II. To quantify the binding of substrates for substrate binding characteristics.
Minimum Specifications:
For biological applications where volume and concentration are limited, we require an instrument with superior sensitivity and minimal sample requirements. Specifically, a system capable of handling 40 µL or less of titrant and the capability of delivering injection volumes as low as 0.1 uL is required. Due to large numbers of samples, we also require the ability to upgrade to an automated system than can handle 100-200 samples a day. Since our users will have varied backgrounds in data fitting, we also require data analysis software allowing for selectable binding models, automated calibration procedures and standardized calibration report generators.
