Description The National Cancer Institute (NCI), Division of Cancer Epidemiology and Genetics (DCEG), Occupational and Environmental Epidemiology Branch (OEEB) plans to procure on a sole source basis services from Vanderbilt University, 2220 Pierce Avenue, 777 Preston Research Building, Nashville, TN 37232-6307 to determine the drive mutations of 40 tumor samples from lung cancer patients in Xuanwei, China. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1). The North American Industry Classification System Code is 621511 and the business size standard is $13.5M.  Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract.  Period of Performance: Performance shall be for twelve (12) months from date of award.  The Division of Cancer Epidemiology and Genetics (DCEG) is an intramural research program of the National Cancer Institute (NCI), National Institutes of Health (NIH) that conducts population and multidisciplinary research to discover the genetic and environmental determinants of cancer and new approaches to cancer prevention.   The contractor shall evaluate driver mutations in tumor samples collected from a study of nonsmoking lung cancer among females in Xuanwei, China. The lung cancer rate among nonsmoking females in this population is among the highest in the world and has been attributed to indoor air pollution from domestic fuel combustion, particularly from smoky coal. This effort will produce new insights into the pathogenesis of lung cancer among nonsmoking females exposed to indoor air pollution from coal, and serve as a comparison with data from lung cancer tumors from nonsmoking females without this exposure in Asia and throughout the world.  The Contractor shall detect 38 recurrent point mutations in 9 genes (EGFR, KRAS, BRAF, NRAS, PIK3CA, MEK1, AKT1, HER2, and PTEN) in tumor tissues and use a PCR-based sizing technique that simultaneously assesses tumors for recurrent insertions in EGFR and HER2 and deletions in EGFR in tumor tissues.  Vanderbilt University and the laboratory of Dr. Pao is the only known source to the NCI that has efficiently and accurately measured driver mutations using methods in tumor samples from subjects in Asia. Compared to direct sequencing, their assays offer higher analytical sensitivity and reduced complexity. They also provide a robust and accessible approach for the rapid identification of important mutations in lung cancer. The data generated must be comparable and have the ability to be pooled with data from previous efforts evaluating driver mutation spectrum among Asian lung cancer populations.
