Chemical Reagents for 384 Genotyping / Multiplex SNP

U.S. Department of Agriculture, Agriculture Research Service, Plant Science Research Unit, Raleigh, NC intends to award a sole source contract to Illumina, Inc., 9885 Towne Centre Dr., San Diego, CA. The Plant Science Research Unit requires reagents for 384 multiplex SNP detection. The equipment would be used to gather SNP data for large numbers of DNA samples in conjunction with the Plant Science Research Unit's genetic investigation of complex traits of plants. In order to determine perform marker-assisted breeding, detection of 384 SNP for each of 1536 barley samples are required.

Reagents requirements:

Must be compatible for use on an Illumina BeadXpress instrument capable of performing barley SNP genotyping and multiplexing for 384 SNP per sample. Must have ability to genotype samples of low quality genomic DNA isolated using SDS protocol. Must be compatible with previously identified, standardized assay conditions requiring no assay optimization and permitting rapid establishment of new assays without redevelopment. Must be able to detect small insertions and deletions in addition to SNPs with the standardized assay. Must have demonstrated ability to do SNP genotyping for complex genome such as barley and wheat.

This research laboratory has an Illumina BeadExpress currently being used in their research which is based on Illumina, Inc., technology; therefore, ordering from another source would compromise the experiments and the integrity of data. The genotyping assay developed by Illumina allows 384 different DNA markers to be analyzed in 96 individuals from a single reaction assay. This high throughput genotyping method permits generating a large amount of data in a short period of time. These chemicals have to be purchased according to the work schedule to maintain their reactivity due to limited shelf life.

This notice of intent is not a request for proposals. The government intends to award this to Illumina, Inc. No other kits are compatible with this equipment. The anticipated award date is July 28, 2011.
