In our studies, the lab routinely separate samples on sucrose gradients. Following centrifugation, the samples are unloaded using an old (>25 years old) ISCO fractionator connected to an analog chart recorder. Despite its age, the system works; however, it has several drawbacks. Most prominently, the use of an analog chart recorder limits the utility of the instrument. We cannot easily quantify the size of peaks, compare results from different experiments, or prepare high quality images for publications. Moreover, our current fractionator is obsolete and spare parts are no longer available. Therefore, when the current system breaks, we must search extensively for replacement parts. The BioComp system corrects these deficiencies.
Use of the BioComp system's digital recorder enables quantification and easy comparison of results from multiple experiments. Because of this advantage, some members of our lab have been carrying fragile gradients across campus to fractionate on another lab's BioComp system. Having a system in our own lab will greatly improve our access to the item and eliminate potential sources of error associated with walking across campus with the samples. Of added benefit, the BioComp system enables fractionation of fluorescently labeled samples - a procedure that we cannot currently perform.
The Biocomp Gradient Fractionator is the only known (to the best of our knowledge) gradient fractionator system that utilizes a digital recorder - enabling quantification, easy comparison of results from multiple experiments, and preparation of high-quality images for publications. The Biocomp system also enables fractionation of fluorescently labeled samples - a procedure that we cannot currently perform and is not accessible on any other fractionator platform. The Biocomp Gradient Fractionator is specifically requested/required as this system is compatible with an existing gradient maker in our lab.
