BRAND NAME OR EQUAL SALIENT CHARACTERISTICS:

In order to optimize the new PCR amplification and sequencing techniques required
for AFDIL's applications, Beckman Coulter Inc. reagents are required. These reagents will result in less re-amplifications being required, conserving both sample and cost to the government.
These improvements will allow for automation, reduction of analyst error, and reduction of overall processing time. During proof of concept and development of the next generation amplification and sequencing strategies, these reagents have been proven to the government to be: reproducible from lot to lot; robust; reliable; and free from human nuclear and mitochondrial DNA contamination. The vendor's ability to utilize quality controls that produce contamination free, reproducible and reliable libraries is essential for processing forensic casework samples and meets AFDIL mission requirements. Development and optimization of the new methodologies to identify missing individuals were performed using these reagents. These new methods are currently being forensically validated in accordance with the Federal Bureau of Investigations Quality Assurance Standards, which has resulted in the need for these items.
Agencourt AMPure XP System 60ml (ManfllA63881); A PCR purification system that utilizes Solid Phase Reversible Immobilization (SPRI) magnetic bead-based technology for post-PCR clean up with no salt carryover. Requires no centrifugation or filtration; can be used in manual and automated 96- well formats. Must be compatible and validated for use with specific Next Generation Sequencing library preparation kits and instruments.

• Must utilize Solid Phase Reversible Immobilization (SPRI) magnetic bead-based technology for post-PCR clean up.
• Must NOT require centrifugation or filtration.
• Must allow for manual and automated processing in both tube and 96-well plate formats.
• Must enable high recovery of PCR amplicons as well as both double and single stranded DNA templates.
