The USDA is looking for an equipment to extract good quality genomic DNA from hard to lyse fungal cells. In our previous studies we have used various types of tissue lyser's and shredders, for sample preparation, however, these were not very effective in extracting good quality DNA from conidia of powdery mildew pathogen of cucurbits and sporangia of the downy mildew pathogen. The system should be easy to use and must allow for rapid and reproducible preparation of the samples to be used for extraction of good quality DNA. The system should use cycling hydrostatic pressure to help lyse or shred the samples. A combination of pressure and temperature to lyse the cells will work too. The system should not be based on the use of glass or ceramic beads to shred the pathogen cells. USDA is interested in doing a rental for three months with the option to buy.
